Cytek Aurora Training Part 1: Introduction to Full Spectrum Flow Cytometry

hello my name is laura johnston i am the scientific associate director at the flow core at the university of chicago now before i get into these videos i want to make two notes first of all i have no affiliation with scitec i created these videos all on my own i did however send a previous version of these videos to scitec to ask for their permission to post them on youtube and they have kindly allowed me to do so second is if you are planning on using our facility at the university of chicago and you have not yet contacted me to schedule your training i would recommend that you stop watching these videos and contact me first however if you have gone through my training and you're just watching these videos as a refresher then you are welcome to proceed now this is the full course outline that i go through in my in-person training but for youtube i will break this down into smaller videos the first thing we'll talk about is what is full spectrum flow cytometry how is it different than our conventional flow cytometers bring the best reference controls this is very important i'll actually have two videos on panel design in my typical course i just go over the tools for panel design so have a video on that but i will also have a second video which is more extensive and i'll walk through a step-by-step panel design process then i have some notes on how to do titrations and some things you need to know about staining conditions to prepare your samples for the aurora we'll talk a bit about the software which is called spectroflow and how spectral flow does the spectral unmixing and then we'll talk about how to check the unmixing accuracy so what is full spectrum flow cytometry and how is it different than conventional flow cytometry so the aurora is a full spectrum flow cytometer and the other cytometers in our facility like the four tesses or the etune those are conventional or classical traditional like to use now full spectrum flow cytometry is a lot of words to say and admittedly i will abbreviate that oftentimes just to spectral cytometry or spectral um but know that the correct term is full spectrum flow cytometry just to remind you of how our conventional flow cytometers are
